I am investigating mechanisms of protein trafficking within the cell and how they contribute to the formation and maturation of lysosome related organelles (LROs) such as WPBs in endothelial cells (ECs). For this I use gene editing techniques (CRISPR/Cas9, shRNA, siRNA) in mammalian cells as model systems to study the complex pathway of protein delivery to subcellular organelles. Currectly, my efforts are focused on VWF trafficking in the context of VWD and how mutations and changes in glycosylation of VWF affect the biogenesis and secretion of WPBs.
