PB1539 - The Influence of G169IA Mutation in the Factor V Gene (Leiden)

Background: In recent years’ studies pointing to the important role of genetic factors as the role of genetic mutation G169IA factor V gene (Leiden) on the development of preeclampsia.

Aims: We have studied the effect of G169IA mutation in the factor V gene (Leiden) on the severity of preeclampsia. For this purpose, 92 pregnant women were examined.
62 of them – with preeclampsia of varying severity and 28 – with uncomplicated pregnancy. Out of 62 pregnant women with preeclampsia 44 had mild preeclampsia, 26 had severe preeclampsia

Methods: The study of the effect of the mutation was carried out by the allele-specific polymerase chain reaction method. The results were confirmed by sequencing – direct determination of the nucleotide sequence of a genomic DNA fragment in the area of the supposed mutation.

Results: In normal pregnancy G169IA mutations in the factor V gene (Leiden) were detected in 24,5% of cases. In all cases it was present in the heterozygous form. The mutation was more common in the group with preeclampsia – in 61% of cases (p = 0,001). There was correlation between the frequency of the mutation and the severity of preeclampsia, which amounted to 55.4% in women with mild and 71.2% - with severe preeclampsia. In women with repeated preeclampsia mutation was detected more often than in primigravida with preeclampsia (84.7% compared to 61.2%, p – less than 0.05).

Conclusion(s): Women with G169IA mutations in the factor V gene have increased risk of developing severe forms of preeclampsia during pregnancy.
